For MT with Cu2+, a model protein with PDB accession code 1JJD was downloaded and used as a representative for cartoon of MT- metal complex. For Apo-MT, the beta-domain of mouse MT-1 with PDB code 1DFT, was downloaded and metal ions are removed using photoshop to represent as a cartoon of protein domain without metal ions. PCA-Nspe does not have any interaction with other bio-metals, e.g.; Ca2+, Mg2+, K+ and Na+ as evident by UV/Vis study (see Supporting Information, Figure S8).
Fe3+ was not added in the list as it increases the absorbance intensity due to pristine absorbance of Fe3+ salt (Figure S8e and ref. [5]), which is avoided for clear interpretation of the signal.

We repeated this separate titration exactly up to 10 equiv because above 10 equiv there might be some free Cu2+ ions in solution that could bind the peptoid chelator upon its addition to the solution mixture giving rise to a false positive signal at 333 nm.
