Chemokine ligands and their cognate receptors play a major role in regulating inflammatory response. Binding to the corresponding chemokine ligand, chemokine receptors can stimulate immune activation and inhibit or promote signaling pathways. CC chemokine receptor 1(CCR1) is a widely studied G-protein-linked receptor target, which is mainly expressed in a variety of leukocytes and is regarded as a good target for autoinflammatory therapy. In addition, CCR1 plays an important role in monocyte extravasation and transport under inflammatory conditions, and CCR1 and its ligands have been extensively studied in a range of inflammatory diseases of the central nervous system (CNS), including multiple sclerosis, Alzheimer's disease, and stroke. The implications have been suggested by animal models, where inhibition of CCR1 or its ligands has shown beneficial effects. This review summarizes the role of CCR1 in inflammatory diseases of the central nervous system, the molecules involved in the inflammatory pathway, and the corresponding drugs or molecules that act after CNS diseases. This evidence may provide possible targets or new strategies for the treatment of inflammatory diseases of the CNS.
